測定原理：
微生物在內的所有活細胞都以ATP作為能量來源，因此，在使用試劑盒中的ATP釋放試劑從微生物中提取ATP後，螢光試劑含有螢火蟲螢光素和螢光素酶，螢光素酶會與ATP反應，透過測量發光量確定微生物數量。
生物發光量與樣品中的ATP能量成正比。
